The singers: David and the captains of the host separate the sons of Asaph, Heman, and Jeduthun, 'who should prophesy with harps, with psalteries, and with cymbals' — prophecy here is a musical office, under fathers and king: Asaph 'prophesied according to the direction of the king.' Then David separated for the service to sing the songs. the sons of Asaph, etc. Out of all the Levites he separated only the sons of Asaph, etc. Heman, and Jeduthun, who prophesied When they would play these musical instruments, they would prophesy. A similar incident [is told] of Elisha: (II Kings 3:15): “’And now fetch me a musician.’ And when the musician played, the hand of the Lord came upon him.” and the number of those men expert in their work... was: i.e., There were twenty- four watches, corresponding to the twenty-four watches [of the priests (Moharich)]: viz. the four sons of Asaph, the six of Jeduthun, and the fourteen of Heman, totaling twenty-four. Below, he sums them all up, as it is written (verse 7): “And their number with their brethren, etc.”

It is possible that the intent here is not that they were prophets, but rather that they would speak by means of harps, psalteries, and cymbals in giving thanks to Hashem, may He be blessed; but the first explanation is more correct.

'and the number of those who did the work according to their service was' — their number matched the number of men required for their particular work: for the vocal song, twelve Levites had to stand on the platform, and for the instrumental accompaniment, the law is that there must be no fewer than two psalteries, nine harps, and one cymbal — which likewise total twelve, as is explained in tractate Arachin and in Rambam's Laws of the Sanctuary Vessels.
